Is it possible to cancel a reservation after a completed online check in?
Not certain what the policy of your reservation is, but yes you can cancel. The question is will you have to pay ?
Since I check on Friday am for a Sunday night room, with travel issues I have had to cancel/change my reservation after check-in. My reservations are all free ot cancel until 6 pm day of check-in (corp rate dictates this).

I have a reservation for Friday and can cancel for free until Thursday. I checked in today Wednesday two days before arrival as I usually do and then realized that I might have to change plans. Just never thought of it before. I’Il gve them a call if necessary.
This is something I've wondered about as well.
My assumption was that once you've checked in online you've lost the right to cancel penalty free, even if you do decide to cancel before the cancellation deadline on your reservation. I can't find any explicit statement on what the implications of checking in (by any method) are, other than a statement that the hotel will reserve credit on your credit card for the cost of the stay when you check-in.
Yes, well, that was my assumption too. Never been in the situation before, could be useful to know if you have to cancel close to arrival.
I've cancelled the day of... Just called the hotel and explained the sitch. Being platinum has always helped. They usually mention it when they say, 'no problem.' The fact that you already checked-in using the mobile app shouldn't make a difference. Make sure you get a cancellation # and the name of the person you spoke with. Now for the disclaimer (all mine). Marriott has fallen off the wagon since the merge, so I wouldn't put anything past them right now and in the foreseeable future.
One time I checked in on the app, then cancelled online the morning of arrival. (I was feeling sick, but tried holding out to see if I’d feel good enough to travel). The property manager actually called me to make sure I had intended on canceling, because she was confused to see a checkin, then a cancelled reservation.
As an FYI, I’ve never had to pay the cancellation fee when canceling after the deadline.
I ALWAYS call the property directly now to cancel, in order to explain the situation (illness, weather, airline). Sometimes I’ve asked if they’d waive the penalty, but most of the time I haven’t needed to; they told me they wouldn’t charge anything because I was Platinum (now Platinum Premier). On one other occasion, they wouldn’t refund my deposit, but they did let me apply it towards a future stay!
I had canceled a room at midnight due to travel issues (stuck in Chicago), being LTP/PP (back then) I didn't have to pay a cancellation fee either.
